The dynamics of structural and functional disorders in rats’ liver in experimental cirrhosis

Abstract.
Objectives. To study the dynamics of structural and functional disorders in the liver of rats with experimental cirrhosis.
Material and methods. Toxic liver cirrhosis in white rats was modelled by prolonged intragastric administration of 40% oil solution of CCl4 during 19 weeks in combination with ethanol. To study the dynamics of structural and functional changes in the liver of experimental animals they were taken from the experiment at different terms: in 3, 6, 9, 12, 16, 19 weeks after the beginning of the experiment. For morphometric evaluation of the content of  lipids succinate dehydrogenase, cytochrome oxidase and connective tissue semi-quantitative method of computer analysis of the results was used. The measurement of the average area of hepatocytes and nuclei (in µm2) was taken with subsequent calculation of the nuclear-cytoplasmic ratio.
Results. As a result of cytomorphometric research the complex dynamics of the nuclear-cytoplasmic ratio change, a significant increase in the average area of hepatocytes and their nuclei and a smooth process of sclerotization of the liver were determined. It should be noted that this study enabled the revealing of statistically significant sex differences in the expression of fatty degeneration after 6, 9, 12, 16 and 19 weeks of the experiment. Steatosis was more clearly marked in females. However, sex differences of enzyme activity were absent. On histochemical study of succinate dehydrogenase mosaic decrease in enzyme activity was noted, it being not observed in cytochrome oxidase. The reduction of enzyme histochemical reaction level testifies to the disturbance of energy supply processes of  hepatocytes in case of cirrhotic process development.
Conclusions. Thus, the conducted study is built on the principle of complex analysis of histological and histochemical indices which reflect the toxic damage of white rats liver. The obtained data reveal some aspects of structural and functional changes in case of the development of toxic cirrhosis of the liver in animals and can be used in the search for ways to correct them.
